So when I say
나보다 더 천천히 먹는 사람이 없는 줄 알았어.
it means "I thought there was no one who eats slower than I do."
Now if I want to say "I knew there was no one who eats slower than I do." how do I say that

The way you constructed it, it means that you held a belief that turned out to be false.
If, in fact there is is no one who eats slower than you, and you knew it, then you would say:
나보다 더 천천히 먹는 사람이 없는 걸 알았어.

hmm... so the only difference is the 줄 or 걸??!

Heh, don't worry, while I can't think of any other case where that 줄 is used (besides the negative of that same construction), that 걸 is actually a contraction for 것을, and you can use that in a lot of other contexts.
That's "...사람이 없는 것을 알았어요."
A verb stem plus -는 것 means "the fact that [verb]" or just "that [verb]". One common example of this in everyday speech is "뭘 하는 거야?" - "What is it that you're doing?" or "어디 가는 거야?" - "Where is it that you're going".

Good question!
-ㄹ지 알다 means “to know whether to/whether it will/etc…”
and
-ㄹ 줄 알다 means “to know how to…”or “to know that it would …”
So sometime the usage for these two patterns are interchangeable, but not always, if the second pattern has the meaning of “to know how to”
Hope this helps!! ^^
